Shan Masood remained unbeaten on 88 off 137 balls with 11 fours, in a 155-run stand with Imam-ul-Haq, who scored 63.
The innings began poorly for Pakistan, with opener Azan Awais bowled by Kemar Roach for a nine-ball duck.
Salman Ali Agha was unbeaten on nought off 9 balls at the close of play.
West Indies had been dismissed for 311 in their first innings, with Mohammad Ali taking four wickets for Pakistan.
Pakistan resume their innings on day three, still trailing by 112 runs.
